摘要
在Triton X-100存在下,红色染料硝基磺酚C与无色血清白蛋白在pH2.5~3.5的酸性介质中生成可溶性蓝色缔合物,其最大吸收波长为683nm,比试剂本身红移128nm,摩尔吸光系数为2.7×105L·mol-1·cm-1,线性范围为0~140mg/L。探讨了反应的最佳条件及初步反应机理。方法可用于临床中尿液总蛋白和脑脊液白蛋白的定量测定。
In the presence of Triton X-100 (TX-100), red Nitrosulfonphenol C(NC) can form a blue complex with human serum albumin (HAS) at pH 2.5 - 3.5. The maximum absorption wavelength is 683 nm, and the molar ab-sorptivity ( ε ) is 2.7 ×10^5 L. mol^-1.cm^-1 . The linear range is between 0 and 140 mg/L. The optimum reaction conditions and the preliminary reaction mechanism are studied. The method has been applied to determination of the proteins in urine samples and cerebrospinal fluid with satisfactory results.
