摘要 目的观察阿魏酸和芍药苷混合剂对FeSO4和H2O2诱导的PC12细胞损伤的影响,初步探讨其作用机制。方法采用FeSO4和H2O2作用产生自由基的方法诱导建立PC12细胞损伤模型。倒置相差显微镜进行细胞形态学观察;采用MTT比色分析测定细胞存活率;硫代巴比妥酸法测定丙二醛(MDA)含量;黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活性。结果与FeSO4和H2O2损伤组进行比较,阿魏酸和芍药苷的混合剂能明显改善PC12细胞的存活数量,降低MDA的含量和提高SOD活性。结论浓度为5~50μmol/L的阿魏酸和浓度为1~10μmol/L的芍药苷混合剂对FeSO4和H2O2诱导PC12细胞损伤有明显的保护作用,其作用机制可能与阿魏酸和芍药苷清除自由基和抑制脂质过氧化有关。 Objective To observe the protective effects of the mixture of ferulic acid (AF) and paeoniflorin on PC12 cells injury by FeSO4/H2O2 in vitro, and explorer its mechanism. Methods The cell injury model was established with FeSO4/H2O2 for 24 h. The process of the growth and morphology of the cells were observed under phase contrast microscope in vitro. The survival rate of the cells was determined by MTT method. The content of MDA and SOD activity was measured respectively by thiobarbituric acid and xanthine oxidase method. Results Compared with FeSO4/H2O2, the mixture of ferulic acid (AF) and paeoniflorin enhanced the PC 12 cells survival rate and viability, reduced MDA content and increased SOD activity. Conclusion The mixture of 5-50μmol/L AF and 1-10 μmol/L paeoniflorin exerted protective effects on PC12 cells injury induced by FeSO4/H2O2, which maybe related to free radical scavenging activity and lipid peroxidation process being inhibited.
