摘要
用逆转录酶/多聚酶链式反应(RT/PCR)技术检测腱抗原蛋白(Tenascin)mRNA在正常角质细胞和良、恶性角质细胞株(HaCaT、SCLⅠ、SCLⅡ)中的表达。结果表明腱抗原蛋白mRNA在正常角质细胞不表达,恶性角质细胞株中高度表达,扩增条带为1300bp,而良性角质细胞株具有弱表达。认为部分细胞有潜在的高增殖能力,可能与细胞生长特性改变有关。
The present study is designed to detect Tenascin(Tn)gene expression in normal keratinocyte,spontaneously transformed human skin keratinocyte (HaCaT) and skin tumor cell lines from squamous cell carcinomas(SCL I,SCL II)by RT/PCR.Our data showed that no expression of TnmRNA in normal keraitnocyte was found, but in tumor cells the Tn gene was distinctly expressed (1300bp).However,weak expression was observed in HaCaT cell line.It is considered that a part of HaCaT cells possesses potentially high cell proliferation and it may be concerned with a change of cell growth character.
