摘要
目的探讨固定剂1(A)、固定剂2(B)、固定剂3(C)、防腐剂1(D)和防腐剂2(E)不同种类的固定剂和防腐剂及不同固定浓度对细菌保存的影响。方法采用正交试验安排试验以变异系数为指标进行固定剂混悬液最优配方的筛选。结果优化的固定剂配方最优组合为A1B3C1D1E1。结论不同的固定剂在不同的固定时间对细菌的保存有显著的影响。优化的固定剂配方对细菌有良好的保存作用,该技术对于尿沉渣细菌的质控物的研制有重要应用价值。
Objective To investigate the effects of different fixed agents (aldehydes, spirits, thymol, and preservatives) and concentrations on bacteria preservation. Methods Optimal compounding of fixative suspension was determined with the orthogonal design and variation coefficient. Results The optimal composition of fixatives was AIB3C1D1E1. Conclusion Bacteria's conservation is markedly affected by different kinds of fixatives and different fixation time. The optimization of fixatives' reagent is good for bacteria's conservation. The technique has its applicable value for preparation of bacterial quality control material of urinary sediment.
