摘要
目的:观察透明质酸钠滴眼液对角膜及隐形眼镜的保护和润滑作用.方法:选用正常新鲜人脐带为原料,经蛋白水解酶酶解消化,氯代十六烷基吡啶和乙醇逐级沉淀,获得透明质酸粗品,精制成钠盐成品,理化分析及质量标准控制达到眼科外用要求,按照眼药配制标准,制成滴眼液.用家兔进行局部致炎反应及毒副作用实验;并对隐形眼镜进行离体药物浸泡试验及动物戴镜观察,将浸泡后的镜片用SEM及相位差显微镜观察其透氧孔;组织学检查戴镜后的兔眼角膜.结果:经动物局部致炎及毒副作用实验证明,无致炎性及毒副作用.应用本滴眼液浸泡的隐形眼镜,SEM及相位差显微镜观察,其镜片透氧孔清晰.动物戴镜72h后,兔眼角膜荧光素染色阴性;组织学检查,角膜上皮细胞完整.结论:透明质酸钠滴眼液作为眼科辅助剂可以用于临床,并对角膜及隐形眼镜具有保护和润滑作用.
Aim: In order to observe the protection and lubricatoin of the corneas and softlens by the eyedrops prepared with hyaluronic acid(HA) Methods: Coarse hyaluronic acid was extracted from normal humanfresh umbilical cord by trypsin digestion and precipitated by cetylpyridium chlorde and ethanol respectively Refined sample was prepared as sodium salt The qualities reached the standard for ophthalmologic clinical use The eyedrops were prepared according to ophthalmologic use Softlens were inundated in the eyedrops and worn in rabbits for 72 hours Holes for oxygen penetration of inundated softlens and corneal histology of rabbits were observed by SEM and microscope phase Results:The eyedrops were proved to be free of inflammatory, toxic andside effects in animal experiments SEM and microscope phase results showed that holes for oxygen penetration of inundated softlens were clear. Staining of corneas by fluorescein was positive Epithelium of the corneas was complete Conclusion: The eyedrops prepared with HA mars sheild and lubricate the corneas and softlens, providing subservient medium for softlens worn clinically.
