利用基因芯片技术研究FGFR3在小鼠胚胎肢芽不同发育阶段的表达 被引量:2

摘要 目的检测在软骨发育分化过程中具有重要作用的成纤维细胞生长因子受体3(FGFR3)在小鼠胚胎肢芽发育过程中的表达。方法采用小鼠全基因组Affymetrix mouse4302.0芯片检测小鼠胚胎肢芽发育不同阶段的基因表达,并进行组织学染色观察。结果FGFR3在E12.5d开始呈现明显的表达上调,在E13.5d达到上调顶峰,E14.5d后FGFR3表达逐渐下调。结论FGFR3的表达与胚胎肢芽软骨发育过程密切相关,在软骨的发育分化过程中发挥着重要的作用,并可作为前软骨细胞的特异性标记物。 Objective To study the expression pattern of fibroblast growth factor receptor 3 (FGFR3) in mouse embryonal limb bud development. Methods Affymetrix mouse 430 2.0 cDNA was used with microarray technique to examine the 34 000 genes. Histology staining observation was performed at the same time. Results FGFR3 expression was enhanced obviously in mouse E12.Sd embryonal limb bud. It continued to increase and peaked in mouse E13.Sd embryonal limb bud. Furthermore, the expression down-regulated gradually from El4.5d in mouse embryonal limb bud. Conclusion The expression of FGFR3 has close correlation with chondrogenesis of mouse embryonal limb bud. FGFR3 may play an important role in chondrogenesis.
