摘要
目的:探讨加味二仙汤对去势大鼠骨质疏松症的防治作用,并揭示其作用机制。方法:成年雌性SD大鼠100只,随机分为五组,分别为正常对照组、病理模型组、雌激素治疗组、加味二仙汤剂治疗组、加味二仙汤颗粒剂治疗组,采取切除双侧卵巢进行骨质疏松症造模,分别给药喂养,在4、13周后每组各取10只大鼠处死,摘取第五腰椎经处理后,分别观察骨组织形态计量变化。结果:给药4、13周后,雌激素治疗组与病理模型组相比骨小梁面积百分比、骨小梁厚度、骨小梁间距均有统计学意义,表现出一定骨保护作用。给药4周、13周后加味二仙汤治疗组与病理模型组相比骨小梁面积百分比、骨小梁厚度、骨小梁间距均有统计学意义,颗粒剂治疗组与汤剂治疗组对比,无显著差异。结论:加味二仙汤能明显改善去势大鼠骨形态计量学指标,其机理是通过剌激成骨细胞生长,抑制破骨细胞活性,并抑制高转换型骨代谢趋势来实现。加味二仙汤颗粒剂防治女性绝经后骨质疏松症具有较高推广价值。
Objective：To research the therapeutic effects of Jiaweierxian granule on osteoporosis in ovariectomized rats and its mechanism. Methods ：100 grown femal SD rats, were randomized into 5 groups ： normal contrast group, pathological model group, estrogen group （ treatment group ） ,jiaweierxian decoction group （ treatment group ）, jiaweierxian granule group （ treatment group ）. All rats were ovariectomized and the osteoporosis model was established. They are fed with medicine respectively, after 4 and 13 weeks, 10 rats in each group were sacrificed to explore. The 5th lumbar vertebrae were cut and stained for bone histomorphometric analysis. Results：After 4 and 13 weeks, estrogen group, compared with pathological model group, showed statistical significance in area percentage, thickness and space of Tb. Jiaweierxian decoction group showed statistical significance compared with pathological model group in area percentage,thickness and space of Tb. Jiaweierxian granule group showed no statistical significance compared with jiaweierxian decoction group. Conclusions：Jiaweierxian decoction can effectively improve the histomorphometric index ,The mechanism may be stimulating osteogenesis and suppressing the cytoactive of osteoclast and trend of the high bone transformation. Jiaweierxian granule deserve wide application in clinical therapy for female osteoporosis after menopause.
