摘要
应用膜片钳全细胞法,研究豚鼠单个心室肌细胞的钠离子通道电流(INa)及其动力学特征。研究显示:INa具有电压和时间依赖性,INa在-50mV激活,在+40mV反转。钠通道动力学包括稳态灭活曲线、使用依赖性阻滞及钠通道灭活后的恢复过程,对其特点的初步讨论表明,Ⅰ类抗心律失常药物与受体亲和力、失活态钠通道受体解离的时间常数,以及其使灭活曲线向左移的电位不同,可以作为Ⅰ类药物亚类的分类基础。
Sodium channel current (INa) and kinetics were investigated using patch clamp whole-cell recording in isolated single ventricular cells from adult guinea pig hearts.The study showed that INa was voltaged- and time-dependent,which was activated at-50mV and reversed at +40mV.The kinetics of sodium channel,including steady-state inactivation,use-dependent block and the time course of recovery from inactivated sodium channel,were discussed and analysed.The study also suggested that the differences of class Ⅰ antiarrhythmic drugs in receptor affinity,time constants for the dissociation from the inactivated receptor as well as potential shift of the inactivated curve coluld be used to subdivide these drugs.
