摘要
TS99是由杀真菌链霉菌产生的一种新型抗真菌农用抗生素。为了明确其适宜的分离纯化工艺,研究了大孔树脂吸附、硅胶柱层析和薄层层析等色谱法分离纯化农抗TS99的条件和效果,结果表明,中等极性的大孔树脂和硅胶是吸附法和色谱法分离纯化TS99的适宜填充剂,其最佳洗脱剂分别为70%甲醇、乙醇∶氨水∶水(8∶1∶1,V/V/V)和展层剂氯仿∶乙醇∶水(2∶3∶1,V/V/V)。发酵液经过预处理后,依次用大孔树脂吸附,硅胶柱吸附层析和薄层层析等分离方法即可获得农用抗生素TS99纯品。
TS99 is a novel antifungal agricultural antibiotic produced by Streptomyces fungicidicus YH9407. In order to choose an economical separation and purification technique for the agricultural antibiotic TS99, conditions and effect of middle polarity macroreticular, silica gel column chromatography and thin-layer chromatography were investigated. Results showed that the macroreticular adsorbent and silica gel were the optimum filler to extract antifungal agricultural antibiotic TS99, and the optimum eluant were 70% methanol, ethanol:ammonia:water(8 : 1 : 1,V/ V/V) and chloroform : ethanol: water (2 : 3 : 1,V/V/V), respectively. Pure TS99 could be obtained from pretreatment fermentation broth through macroreticular adsorbent, silica gel column chromatography and thin-layer chromatography.
