摘要
目的建立曲克芦丁的血药浓度测定方法。方法采用反相高效液相色谱法,色谱柱为Hypersoil BDS-C18柱(150mm×5mm,5μm),以甲醇-水-冰乙酸(40:60:5)为流动相,柱温30℃,流速0.8mL/min。以苯甲酸(1mg/mL)作内标,甲醇沉淀蛋白,离心后取上清液进样,于254nm波长检测。结果曲克芦丁质量浓度线性范围是1.0~50μg/mL,r=0.9996(n=8);最低检测限为50ng/mL,高、中、低3种质量浓度平均回收率为97.61%~100.72%,日内和日间精密度RSD分别小于3.38%和4.94%。结论该法简便、快捷,可用于血浆中曲克芦丁的测定。
Objective To establish a RP- HPLC method for the determination of troxerutin concentration in plasma. Methods The samples were determined by RP-HPLC on the Hypersoil BDS-C18 column with the mixture of CH3OH-H2O-CH3COOH(40:60:5) as the mobile phase and the UV detection at 254 nm. Results The linear range was from 1.0-50 μg/mL with r=0.9996(n=8).The detection limit was 50 ng/mL. The average recovery rates with high, middle and low concentrations were 97.61% -100. 72%. The RSD of intra-day and inter- day were 〈 3.38% and 〈 4.94% respectively. Conclusion The method is simple, rapid and stable for the determination of troxerutln concentration in plasma.
