摘要
目的:利用荷瘤动物模型初步探讨化学免疫治疗肺癌时不同程序联合应用对肿瘤生长及T细胞功能的影响,以明确最佳治疗方案。方法:建立C57BL/61Leuwis肺癌荷瘤小鼠动物模型,全身应用卡铂及局部应用TNFα、IFNγ,不同程序联合处理荷瘤鼠,观察小鼠肿瘤生长情况。用同位素掺入法检测脾脏T细胞转化能力及脾细胞对IL-2的应答能力。结果:肿瘤生长速率最小者为先卡铂后细胞因子组(P<0.001);其次为卡铂与细胞因子同时组(P<0.01);T细胞转化能力最高组为卡铂、细胞因子同时组(P<0.001)。其他两组升高明显(P<0.05),脾细胞对IL-2的应答能力以同时治疗组和先细胞因子组最高(P<0.05)。结论:不同程序的联合方案对机体T细胞功能影响不同;卡铂与细胞因子同时应用为最佳治疗方案。
Objective:The growth rate of lung tumor and the changes of T cell functions were studied with experimental tumor-bearing mice after being treated with different chemo-immunotherapy procedure,in order to find out the best scheme. Methods:C57BL/6 mice bearing Lewis lung cancer were used as murine tumor model.The growth rate of tumor were examined after carboplatin was used in whole-body and TNFα、IFNγ were used in different procedure in local-body. The ability of spleen T cell transformation and response to IL-2 were examined with the method of isotopic incorpoation. Results:The smallest percent of the tumor growth was in the carboplatin-first group (P<0. 001 ),and the second one was in the simultaneous group (P<0. 01 ). The highest transformation of spleen T cell was in the simultaneous group (P<0. 001). The other two groups changed obviously (P<0. 05). The highest resPOnse to IL-2 of spleen cell was in the simultaneous and cytokines-first group (P<0.05). Conclusion: The effect on T cell function was fifferent according to different therapy procedure. The scheme of carboplatin and cytokines used simultaneously is the best.
