摘要
应用异型双功能连接剂SPDP将抗人肺腺癌单克隆抗体CMU15A与天花粉蛋白(TCS)交联,制备出新型免疫毒素。应用的MTT法检测其对人肺腺癌细胞系AGZY—83A的选择性杀伤活性。结果表明,CMU15A—TCS结合物具有高效靶向杀伤肺腺癌细胞能力,优于游离TCS及单抗与TCS混合物;与对非靶细胞的细胞毒作用相比较差异非常显著。提示该结合物是一种很有价值的免疫治疗剂,为今后的实验和临床应用提供了资料。
he immunotoxins were prepared by conjugating trichosanthin(TCS)with monoclonal antibodyCMU 15A.Study in vitro indicated that CMU 15A-TCS conjugate retained substantially the original im-munological activity of the CMU15A and showed stronger selective cytotoxicity against human lung ade-nocarcinoma cell line AGZY-83A(IC50 0.91 μg/ml)than free TCS(IC50 82μg/ml)and mixtures ofCMU 15A and TCS(IC50 60μg/ml),The difference was significant in comparison with cytotoxicity a-gainst nontarget cell line(Hela).The result suggested that CMU15A-TCS conjugate was an effective immunotoxin.
