灵芝多糖对肾小球系膜细胞增殖的影响和体外抗氧化活性的研究 被引量:14

Inhibitory Effect of Ganoderma Lucidum Polysaccharide on the High Glucose-Induced Proliferation of Human Mesangial Cells and Antioxidant Activity in Vitro

摘要 采用纤维素酶法提取灵芝多糖,获得两个组分cGLP1和cGLP2,以MTT法测定两组分对高糖诱导人肾小球系膜细胞(HMC,human Mesangial Cell)增殖的影响,并通过对O2.-、.OH两种自由基的清除作用以及总抗氧化能力来测定其抗氧化活性。进而考查酶法提取的灵芝多糖(Ganoderma Lucidum polysaccharide)对高糖引起肾小球系膜细胞增殖的抑制作用及其体外抗氧化活性。结果显示cGLP2对高糖所致的HMC增殖有显著的抑制作用,且cGLP2对羟自由基具有明显的清除作用,并具有较强的抑制过氧化脂质反应的能力。表明灵芝多糖酶提组分cGLP2能显著抑制高糖诱导的HMC增殖,这可能与cGLP2抗氧化活性相关。 To investigate the inhibitory effect of polysaecharide extracted with cellulose, on the high glucose-induced proliferation of HMC and antioxidant activities in vitro, two kinds of polysaccharides puri fied from Ganoderma Lucidum were named cGLP1 (Mw=4700)and cGLP2 (Mw=1000000). Inhibitory effects of polysaccharide on the high glucose-induced proliferation of HMC was measured by MTT. Antioxidant activities were determined by superoxide and hydroxyl radical scavenging activity, also by linoleic acid oxidation inhibition capability. The results showed that cGLP2 can obviously inhibit the proliferation of HMC induced by high glucose. It also has the obvious antioxidant activity such as hydroxyl radical scavenging and inhibiting of lipid peroxidation, so the inhibitory effect of cGLP2on the high glucose-induced proliferation of HMC may be relative to its antioxidant activity.
