摘要
以8个枣树品种的嫩叶为材料,比较了不同的DNA提取方法,结果表明:采用改良的CTAB法提取的DNA纯度和产率,虽然不比改良SDS的高,但适宜RAPD分析.在20μL反应体积中,RAPD分析的优化反应体系为:引物0.2 pmol.L-1、Mg2+2.0 mmol.L-1、dNTP200μmol.L-1、TaqDNA聚合酶1.2 U.改良的CTAB法是枣树RAPD分析基因组提取DNA的最好方法.
Using the tender leaves of 8 jujube trees as the experimental material, compared with the different methods of genomic DNA extraction, authors studies the method to the genomic DNA extraction and build RAPD system. The results obtained from this research indicate that the purity and productivity of DNA through the modified CTAB is suitable to the RAPD analysis although it is lower than the modified SDS' s. The optimal RAPD analysis was as follows:0.2 pmol · L^-1 random primer, 2.0 mmol · L^-1 Mg^2+ , 200 μmol · L^-1 dNTP, and 1.2 U Taq polymerase in 20 μL reaction solution . The modified CTAB is the best method of genomic DNA extraction when RAPD is used to analyze the genome of jujube.
