摘要
目的:观察人参皂苷Rg1对成年大鼠脑缺血后海马区神经干细胞增殖分化的影响,探讨其促进神经细胞再生的作用.方法:将60只Wistar大鼠随机分为假手术组、单纯缺血组及人参皂苷Rg1治疗组.用Longa线栓法制作大鼠脑中动脉闭塞模型.通过腹腔注射5-溴脱氧尿苷(BrdU)的方法标记神经干细胞,用免疫单标及双标免疫荧光技术观察人参皂苷Rg1对局灶性脑缺血后海马区BrdU,BrdU/β-Ⅲ型微管蛋白(Tuj-1)和BrdU/波形蛋白(Vimentin)免疫活性的影响,并计数作定量分析.结果:免疫组织化学染色显示脑缺血后海马区存在BrdU阳性细胞.单纯缺血组7d(28.4±3.0)时,与同组1d(12.7±1.2),3d(20.3±2.7)相比较,差异具有统计学意义(P<0.01),且缺血7d后阳性细胞达高峰,14d(17.2±1.2)时可见BrdU阳性细胞急剧减少.应用人皂苷Rg1后BrdU阳性细胞数明显增多,人参皂苷Rg1治疗组1d(15.1±1.5),3d(27.2±3.3),7d(37.1±4.2),14d(23.6±2.7)与同时间点单纯缺血组相比差异均具有统计学意义(P<0.01);人参皂苷Rg1治疗组BrdU/Tuj-1,BrdU/Vimentin阳性细胞数分别为(14.3±0.5),(15.1±0.5)个/视野,较单纯缺血组(7.2±0.3),(8.9±0.4)个/视野明显增多(P<0.01).结论:人参皂苷Rg1能诱导海马区神经干细胞增殖、分化,提示其具有促进神经神经细胞再生的作用.
AIM: To observe the influence of the ginsenoside Rgl on proliferation and differentiation of neural stem cells (NSCs) in the hippocampus zone after cerebral ischemia in adult rats, so as to study the role of the ginsenoside Rgl in promoting neural cell regeneration. METHODS : Sixty Wistar rats were randomized into sham operation group, pure ischemia group and ginsenoside Rgl treatment group. Model of middle cerebral artery occlusion was made by Longa's method. NSCs were labeled by the intraperitoneal injection of Bromodeoxyuridine (BrdU). Immunohistochemical single staining and double-immunofluorescence technique were used to investigate the effects of ginsenoside Rgl on the immune activity of BrdU, BrdU/class Ⅲβ-tubulin( Tuj-1 ) and BrdU/Vimentin in the hippocampus zone after focal cerebral ischemia, and the cells were counted for quantitative analysis. RESULTS: BrdU-positive cells were observed in the hippocampus zone after cerebral ischemia by immunohistochemical staining. Compared with 1 d (12.7±1.2) and 3 d (20.3± 2.7) in the same group, pure ischemia group 7 d (28.4±3.0) showed statistical difference ( P 〈 0.01 ), and the positive cells reached the peak at the 7 th day after ischemia. BrdU-positive cells decreased rapidly at 14 d ( 17.2 ± 1.2). After the treatment of applying ginsenoside Rgl, the expression of BrdU-positive cells increased markedly. Compared with the same time point in pure ischemia group, the ginsenoside Rgl treatment group 1 d (15.1 ±1.5), 3 d (27.2 ±3.3), 7 d (37.1±4.2) and 14 d (23.6 ±2.7 ) showed statistical difference ( P 〈 0.01 ). Compared with the pure ischemia group (7. 2 ± 0. 3 )/field and (8.9±0.4)/field, numbers of BrdU/Tuj-1 and BrdU/Vimentin positive cells were respectively (14.3± 0.5 )/field and (15.1 ± 0.5 )/field in the treatment group and much higher with statistical significance ( P 〈 0.01 ). CONCLUSION : Ginsenoside Rgl can induce the proliferation and differentiation
