基于迭代方法的单细胞凝胶电泳彗星图像自动双阈值分割 被引量:2

Automatic double-threshold segmentation of comet images in single cell gel electrophoresis based on iterative arithmetic

摘要 单细胞凝胶电泳(彗星)试验分析中,彗星图像的自动分析能够快速、准确地获取多个彗星指标,这有助于提高彗星试验的分析速度和试验结果的准确度、可比度。彗星轮廓的合理分割是后续分析各项指标的前提。由于彗星尾部灰度值较低且与背景对比度差,导致彗星轮廓自动分割的困难。本文在分析彗星图像特点和比较现有分割方法的基础上,对传统迭代阈值分割方法进行了改进,并推广至双阈值迭代分割。测试结果表明,迭代双阈值分割方法具有快速、可靠、分割效果理想等优点,可以较好地实现彗星图像的自动分割。 In the analysis of single cell gel electrophoresis ( SCGE), i.e. comet assay ( CA), many reliable parameters of the comet can be obtained rapidly by the comet image automatic analysis system which can improve the accuracy and comparability of the CA. Reasonable segmentation of the comet contour is the precondition for all parameter measurement during the automated analysis for the CA, but it is difficult to extract the comet contour from the comet image because of the lower gray-level in the tail region and the bad contrast between the comet tail and the background. Characters of comet images and the existing segmentation methods for their segmentation are analyzed first, then the traditional iterative method is improved and extended to the double-threshold. By this improved iterative double-threshold method, the segmentation resuits for many comet images are satisfactory, and this improved method can be used for quick and reliable segmentation of the comet images.
