康宁木霉cbh2基因的克隆与结构特征分析 被引量:1

摘要 克隆得到了康宁木霉AS3.2774的cbh2基因组DNA及cDNA,序列测定表明:基因组DNA全长1583 bp,cDNA全长1413 bp。对比cbh2基因组DNA序列与cDNA序列,证明该基因由4个外显子组成,被3个内含子间断隔开,编码470个氨基酸的多肽。利用Predictprotein软件预测,该蛋白由9个α螺旋结构和10个β折叠结构及其他结构组成,并对其纤维素结构区、2个糖苷水解酶家族6的特征结构区、信号肽等特征性结构区进行了定位。 The genome DNA and cDNA of cbh2 gene of Trichoderma Koningii were cloned and sequenced. The sequence analysis showed that the genome DNA and cDNA of cbh2 gene were 1 583hp and 1413bp, respectively. Through comparison, the two DNA sequences showed 4 exons that were separated by 3 introns and could encode a peptide of 470 amino acids. By means of Pre- dietprotein software, the protein had 8 α-helixes,7 β-sheets and some other structures. The locations of cellulose binding domain, 6 conserved domains of two glycosylhydrolase family and signal peptide in the protein were confirmed.
