摘要
目的:明确注射用盐酸环维黄杨星 D 在人体中药代动力学特征。方法:色谱条件:Kromasil CN 柱(150 mm×4.6 mm,5μm);流动相:甲醇-水-甲酸(80:20:0.4);流速1 mL·min^(-1);柱温:25℃。质谱条件:离子源为 ESI 源;喷雾电压:5.0 kV;毛细管温度:350℃;源内碰撞电压:25 V。32名健康受试者,随机分为4组,单次给药3个剂量,多次给药1个剂量组,用 LC-MS/MS 法测定血浆中环维黄杨星 D 的浓度,计算药代动力学参数。结果:单次给药2 mg 组:V1/F:(241.909±91.829)L,CL/F:(52.566±35.007)L·h^(-1),AUC_(0-48):(35.358±13.311)μg·h·L^(-1);4 mg 组:V1/F:(425.243±167.074)L,CL/F:(68.468±32.36)L·h^(-1),AUC_(0~48):(42.316±11.271)μg·h·L^(-1);6 mg 组:V1/F:(408.318±218.488)L,CL/F:(70.211±25.616)L·h^(-1),AUC_(0-48):(78.896±34.031)μg·h·L^(-1);多次给药2 mg 组:V1/F:(66.517±71.233)L,CL/F:(13.925±7.488)L·h^(-1),AUC_(0~48):(82.957±25.821)μg·h·L^(-1)。结论:盐酸环维黄杨星 D 为线性药物,每天给药1次体内未见蓄积,推荐临床在2~6 mg 范围内按每天1次给药。
Objective: To study the pharmacokinetics of cyclovirobuxine D for injenction in healthy volunteers. Method : Kromasil CN column ( 150 mm × 4.6 mm, 5 μm ) , mobile phase : water - fomic acid ( 80: 20 : 0.4 ) , flow rate : 1.0 mL ·min ; column temperature :25 ℃. Ion rood : ESI ; atomization voltage :5.0 kV ; eaillary temperature :350 ℃,collision voltage :25 V. Results: Single -dose 2,4,6 mg and multiple -dose 2 mg of injective cyelovirobuxine D were given to 32 healthy volunteers in randomized crossover study. The concentration of cyclovirobuxine D in plasma was determined by LC - MS/MS. The pharmacokinetic parameters were calculated. The pharmacokinetic parameters cyelovirobuxine D and reference were as follows : single dose 2 mg: V1/F-(241.909±91.829)L,CL/F:(52.566±35.007)L·h-1,AUC0-48:(35.358±13.311)μg·h·L-1;4mg :V1/F:(425.243±167.074)L,CL/F:(68.468±32.36)L·h-1,AUC0~48:(42.316±11.271)μg·h·L-1;6mg:V1/F:(408.318±218.488)L,CL/F:(70.211±25.616)L·h-1,AUC0-48:(78.896±34.031)μg·h·L-1;multiple-dose 2mg:V1/F:(66.517±71.233)L,CL/F:(13.925±7.488)L·h-1,AUC0-48:(82.957±25.821)μg·h·L-1.. Conclusion: Cyelov- irobuxine D for injection is linear drug. When eyclovirobuxine D was given at once - daily doses, it is not cumula- tion. It is commended 2 -6 mg at once -daily doses in clinical application.
