摘要
目的:研究高浓度葡萄糖诱导人LECs凋亡以及胰岛素对LECs凋亡的影响。方法:采用终浓度为5.56,11.11和22.22mmol/L葡萄糖处理体外培养的LECs,诱导建立LECs凋亡模型。用终浓度为11.00μU/mL的胰岛素进行干预。利用Giemsa染色法、流式细胞仪和透射电镜对LECs凋亡情况进行检测。结果:Giemsa染色和透射电镜发现了凋亡的LECs。流式细胞仪检测发现随着葡萄糖浓度增高,LECs凋亡百分率亦呈升高趋势,不同组间差异有显著性。通过胰岛素干预可降低由高糖引起的LECs凋亡百分率。结论:高浓度葡萄糖可在体外诱导兔LECs凋亡。胰岛素可保护LECs并减少其凋亡的发生。
AIM: To observe human lens epithelial cells (LECs) apoptosis induced by high ambient glucose and the effect of insulin on LECs apoptosis. METHODS: LECs cultured in vitro were dealt with various concentrations of high sucrose respectively (5.56, 11.11, 22.22mmol/L). A model of apoptotic LECs was established, and then intervened with 11. 00uU/mL insulin. The apoptosis of LECs were observed by Giemsa staining, flow cytometry(FCM) and transmission electron microscopy(TEM) respectively. RESULTS. The apoptotic of LECs were founded by using Giemsa staining and TEM. Quantified by FCM, the percentage of apoptotic LECs escalated with glucose concentration increasing, and significant differences existed in different groups. The percentage of apoptotic LECs decreased after using insulin. CONCLUSION.. The apoptosis of rat 's LECs can be induced by high ambient glucose in vitro. Insulin is able to protect LECs and reduce the occurrence of LECs apoptosis.
