细胞色素C在结肠癌组织中的表达及其与细胞线粒体超微结构改变的关系 被引量:2

摘要 目的:研究结肠病变时细胞凋亡途径及线粒体在凋亡过程中的作用,以了解其在结肠癌生物治疗中的价值。方法:采用原位分子杂交方法(ISH)检测60例结肠癌(肠癌组)、19例结肠腺瘤(腺瘤组)组织及18例正常肠黏膜组织(正常组)中内源性促凋亡蛋白细胞色素C mRNA的表达。并对其中4例结肠癌、6例结肠腺瘤以及7例正常肠黏膜细胞线粒体进行超微结构观察。结果:细胞色素C在肠癌组、腺瘤组及正常组中的表达阳性率分别为93.3%(56/60例)、73.7%(14/19例)及38.9%(7/18例),肠癌组与正常组及腺瘤组与正常组比较,χ2值分别为27.10、4.56,均P<0.05,差异有统计学意义。肠癌组与腺瘤组比较差异未见统计学意义。肠癌组细胞色素C阳性率与性别、年龄、肿瘤大小、组织学类型及Duckes分期等因素无关,但随着分化程度的升高,阳性表达也逐渐升高;低、中、高分化结肠癌阳性表达率次第为87.5%(14/16例)、93.9%(31/33例)及100%(11/11例)。细胞线粒体透射电镜超微结构观察所见,细胞色素C呈高表达者,肠癌组及正常组均为2/4,腺瘤组为4/6。肠癌组线粒体大小不一,分布不均,基质不均匀,嵴发育不良且分布不均,少数线粒体膜破裂,但内质网结构良好。腺瘤组大部分细胞线粒体丰富,规则,卵圆形,大小较一致,嵴发育良好,但基质不均,少部分线粒体嵴有断裂。正常组线粒体嵴发育良好,基质均匀,少部分线粒体嵴有断裂,粗面内质网丰富。结论:从线粒体释放到细胞浆中的细胞色素C可诱导结肠细胞内源性凋亡,此种凋亡受到抑制,从而引起结肠癌的发生。 Objective To study the intrinsic apoptotic pathway of the colorectal cancer cells and to study the possibility that molecular targeting therapy may be used to treat the eolorectal cancer. Methods Sixty cases of eolorectal cancer specimens (eoloreetal cancer group, CRC group), 19 cases of colorectan adenoma (colorecta adenoma group, CRA group) and 18 cases of eoloreetan nomorl(colorecta nomorl group, CRN group) were examined for the pro-apoptotic cytochrome C by in situ hybridization (ISH) method. The uhramicrostructural changes were studied in 4 cases of colorectal cancer and 6 cases of colorectal adenoma with 7 healthy adult's colorecta tissues as control. The ultramicrostructural examination with special attention to the mitochondrial changes were studied in 4 cases of colorectal cancer and 6 cases of colorectal adenoma seven healthy adult's colorectal tissues as control. Results The positive rates of expression of cytoehrome C were 93.3%(56/60 cases)in CRC group and 73.7%(14/19cases) in CRA group and 38.9% (7/18 eases) in CRN group. There were no statistic difference between them. As intensity of eytoehrome C was divided into (-), (+), (++) and (+++). In this study the positive rate of expression of eytochrom C increased with differential degree of the cancer cells. The positive rate of expression in low medium and high degree of differentiation were respectively 87.5% (14/16 cases), 93.9% (31/33 cases) and 100%(11/11 cases). In mitoehondria uhramicrostructural examination the expression of high intensity of cytochrome C was 2/4 cases both in CRC group and normal control group, 4/6 cases in CRA group. In mitochondria, crista were found deformed in shape and decreased in number, many crista disappeared, some mitochondria with ruptured membranes and herniation of matrix. Conclussion The apoptosis proceeded in the coloreetal cancer is mainly through the intrinsic apoptosis pathway(mitochondria mediated pathway).
