摘要
目的探讨姜黄素对痴呆模型小鼠学习记忆能力的改善作用和对脑细胞凋亡及凋亡相关蛋白的影响。方法选用自然快速脑老化小鼠为研究对象,随机将SAMP8脑老化小鼠分为模型组、姜黄素治疗组和盐酸多奈哌齐对照组,将SAMR1小鼠作为正常对照组。采用行为学方法评价各组小鼠治疗前后的学习记忆能力变化,TUNEL法检测海马与皮层的细胞凋亡率,免疫细胞化学法测定细胞凋亡相关蛋白bcl-2和bax的表达情况。结果姜黄素能够明显改善自然快速脑老化小鼠的学习记忆能力,与模型组比较差异有统计学意义(P<0.05);姜黄素治疗组小鼠海马和皮层的细胞凋亡数减少,与模型组比较差异有统计学意义(P<0.05);凋亡相关蛋白bcl-2表达升高,bax表达降低,并且bcl-2/bax比值升高,差异具有统计学意义(P<0.05)。结论姜黄素对痴呆模型小鼠的学习记忆能力具有一定的改善作用,其作用机制可能与其调节凋亡相关蛋白bcl-2与bax的表达有关。
Aim To observe the effects of curcumin on the ability of learning and memory of senescence-accelerated mouse ( SAMP8 ) , as well as it' s influence on brain cell' s apoptosis and proteins related to apoptosis. Methods Sixty 8-month-old senescence accelerated mice were randomly divided into model group, cur-cumin group and donepezil hydrochloride group. Twenty 8-month-old SAMR1 were used as control group. After being treated for 90 days,we evaluated the mice' s ability of learning and memory by behavior means, the apoptosis of hippocampus and cortex by TUNEL test, and the expression of apoptosis-related protein bcl-2 and bax by immunohistochemical method. Result Compared with the model group,mice in the curcumin group improved the ability of learning and memory (P〈 0. 05 ), with less apoptosis cells ( P 〈 0.05 ), higher expression of bcl-2 ( P 〈 0. 05 ) , lower expression of bax ( P 〈 0.05 ), and higher ratio of bcl-2/bax. Conclusion Curcumin can improve senescence-accelerated mouse's ability of learning and memory, probably because it can modulate the expresion of apoptosis-related protein bcl-2 and bax.
