四氯化碳和醋氨酚急性肝损伤犬模型的比较 被引量:4

摘要 目的:比较四氯化碳(CCl4)和醋氨酚(APAP)所致急性肝损伤犬模型的异同,为药物性肝炎的相关研究提供动物模型选择的实验依据。方法:24只健康杂种犬随机分为CCl4组和APAP组(n=12)。CCl4组一次性腹腔注射50.0%(W/W)CCl4花生油溶液0.9mL/kg,APAP组3次多点皮下注射APAP溶液。分别观察各犬的全身情况、记录生存时间,测定血液生化指标,犬死亡时或存活15d时取肝脏组织行病理检查。结果:两组生存率比较差异无显著性(P>0.05),血液生化指标3d内达高峰;CCl4对肝功能的影响比APAP明显(P<0.05),而APAP肾毒性较CCl4大(P<0.05);CCl4组肝炎表现较重,肝脏病理改变较显著;CCl4组的肝外表现较APAP组显著。结论:CCl4对犬的生存状态影响和肝脏的损害程度均高于APAP,而APAP的肾毒性较CCl4显著。 Objective To investigate the similarities and differences between carbon tetrachloride (CCl4) and aeetaminophen (APAP) induced canine liver failure models, and to supply experimental data for studies of drug-related hepatitis. Methods 24 dogs were randomly divided into CCl4 group and APAP group. Dogs in CCI4 group were injected intraperitoneally with CCl4 mixed with the same amount(50%, W/W) of peanut oil in the dosage of 0.9 mL per kilogram of body weight. Dogs in APAP group were given three subcutaneous injections of APAP in dimethyl sulfoxide (600 mg/mL), with the first dose of 750 mg/kg body weight, the second and third dose of 200 mg/kg body weight separately at 9 h and 24 h after the initial injection. The general condition, survival time, the biochemical and hematological indexes (alanine aminotransferase, aspartate aminotransferase/alanine aminotransferase AST/ALT, serum total bilirubin, prothrombin time, blood ammonia, blood urea nitrogen)were measured. The hepatic pathological changes were observed on 15th d or when the dogs died. Results There was no significant difference between the survival rates of two groups (P 〉 0.05). The biochemical and hematological indexes grew to peak in three days. CCI4 could significantly affect liver function (P 〈 0.05 ), while APAP had more serious renal toxicity(P 〈 0.05 ). Dogs in CCl4 group showed more serious hepatitis than APAP group. Also the hepatic pathological changes were significant in group CCl4 After CCl4 injection, dogs showed significant changes in neural symptom immediately and in mental state and diet in the following days. Conclusion CCl4 could significantly affect the general condition and liver function of dog more seriously than APAP. However APAP had more serious nephrotoxicity than CCl4.
