子宫内膜癌组织中PTEN与VEGF、MVD的相关性分析 被引量:2

摘要 目的研究抑癌基因PTEN、血管内皮生长因子和微血管密度表达改变在子宫内膜癌发生及预后中的作用。方法采用免疫组织化学方法检测抑癌基因PTEN、血管内皮生长因子和微血管密度在51例子宫内膜癌和15例正常子宫内膜组织中的表达;并对51例子宫内膜癌患者进行术后随访。结果51例子宫内膜癌组织中抑癌基因PTEN、血管内皮生长因子表达率和微血管密度,平均值分别为43.1%、76.5%、35.32±10.09,与正常子宫内膜比较均有显著性差异(χ2分别为8.817、19.636,t=34.254,均P<0.05)。抑癌基因PTEN、血管内皮生长因子、微血管密度表达与子宫内膜癌的临床病理特征存在一定的关系。子宫内膜癌生存时间与手术病理分期、组织分级、肌层浸润以及血管内皮生长因子、微血管密度表达有关;但肌层浸润、手术病理分期、微血管密度表达是影响预后的主要危险因素之一(P<0.05)。抑癌基因PTEN对预后影响无统计学意义(P>0.05)。结论血管内皮生长因子、微血管密度高表达和抑癌基因PTEN表达降低可能是参与子宫内膜癌发生发展的重要因素之一。子宫内膜癌的手术病理分期、组织分级、肌层浸润以及血管内皮生长因子、微血管密度表达是评价子宫内膜癌预后的重要指标,但肌层浸润、手术病理分期、微血管密度是重要危险因素。 Objective To investigate relationships of expressions of phosphatase and tensin homologue deleted on chromosome ten(PTEN) vascular endothelial growth factor (VEGF) and microvessel density (MVD) with development and prognosis of endometrial carcinoma. Methods The expressions of anti-oncogen PTEN, VEGF and MVD in 51 cases of endometrial carcinoma and 15 cases of normal endometrial tissues were detected by using immunohistochemical method. All 51 patients with endometrial carcinoma were followed up after operation. Results The positive expression rates of PTEN and VEGF and mean MVD in 51 cases of endometrial carcinoma were 43.1%, 76.5% and 35.32±10. 09 respectively, which were significantly higher than those in the normal endometrial tissues and the differences were significant (χ^2 = 8. 817, 19. 636, respectively, t = 34. 254, all P 〈 0. 05 ). The expressions of PTEN and VEGF and MVD had some relationships with clinical pathological characteristics of the malignancy and there were negative correlations between PTEN and VEGF, PTEN and MVD. The short-term survival time of the patient with endometrial carcinoma was related to postoperative pathological stage, histological grading, invasion to myometrium, expression of VEGF and MVD. The invasion to the myometrium, postoperative pathological staging and MVD were main risk factors to influence prognosis of the patients with endometrial carcinoma ( all P 〈 O. 05). While expression of anti-oncogen PTEN was not associated with prognosis of the patients (P 〉 0.05 ). Conclusion The over-expressions of VEGF and MVD and decreased expression of PTEN may be important factors to promote occurrence and development of endometrial carcinoma. Postoperative pathological stage, histological grading, invasion to myometrium, over-expression of VEGF and MVD were important indexes to evaluate prognosis of the patients with endometrial carcinoma, of which, invasion to myometrium, postoperative pathological stage and MVD are important risk factors of prognosis
