摘要
目的考察不同来源粉葛中总黄酮、葛根素的含量。方法采用紫外分光光度法(UV)测定总黄酮含量;采用高效液相色谱法(HPLC)测定葛根素含量,kromasil C18色谱柱(4.6mm×250nm,5μm),流动相为甲醇-水(33:67),流速为1.0ml·min^-1,柱温30℃,检测波长为250nm。结果总黄酮含量为0.53%~1.03%,回收率为99.69%,RSD〈2%(n=6);葛根素含量为0.21%~0.49%,回收率为96.11%,RSD〈2%(n=6)。结论不同来源的粉葛药材中总黄酮和葛根素含量间变异较大,其中广东产粉葛,其总黄酮和葛根素含量均较高,平均质量优于广西产粉葛。
Objective To determine the content of total flavonoids and puerarin in Radix Puerariae thomsonii of different originations. Methods The content of total flavonoids was determined by ultraviolet spectrophotometry (UV). The content of puerarin was determined by HPLC, the analysis was performed on a kromasil C18 column (4. 6 mm ×250 mm, 51μm) with methanol -water (33:67) as mobile phase at a flow rate of 1. 0 ml · min^- 1, and at a column temperature of 30℃. The detection of wavelength was 250 nm. Results The content of total flavonoids was 0.53 - 1.03%, and recovery rate was 99.69 %, RSD 〈2% (n = 6). The content of puerarin was 0.21% -049% , and recovery rate was 99. 11 % , RSD 〈2% (n = 6) . Conclusion The content of total flavonoids and pucrarin in Radix Puerariae thomsonii has great changes between the different originations, the contents of total flavonoids and puerarin in Guangdong Radix Puerariae thomsonii are higher, and the average quality is better than Guangxi Radix Puerariae thomsonii.
