降低彩色马蹄莲组培初代培养污染因素的筛选 被引量:4

Screening on the Methods for Decreasing Contamination Rate of Tissue Culture for Zantedeschia antedeschia in Initial Culture

摘要 [目的]筛选降低彩色马蹄莲组培初代培养污染率的最佳方法。[方法]以正在萌芽的红色、黄色马蹄莲1年生小球茎为培养材料,选取水浴温度、表面消毒时间、表面消毒剂种类、防腐剂种类4个因素进行正交设计,培养10 d后统计每个组合的污染率、褐变率。[结果]选取的4种因素对污染率均有极显著影响(P<0.01),其中水浴温度、表面灭菌时间、表面消毒剂种类对褐变率有显著影响(P<0.05)。从单个因素来看,水浴温度40℃时污染率较低,0.1%HgC l2处理的污染率较低,无防腐剂处理的污染率较低;水浴温度45℃时褐变率较低,表面灭菌时间短的处理减少褐变率的效果较好。[结论]水浴温度50℃、0.1%HgC l2处理20 m in、无防腐剂培养基的污染率最低。 [ Objective ] The aim was to screen the optimum method for decreasing the contamination rate of tissue culture for Zantedeschia antedeschia in initial culture. [ Method ] With the budding annual bulbet of red and yellow Zantedeschia as the cultured materials, 4 factors of bath temperature, surface disinfection time, kinds of surface disinfectant and preservative were selected for orthogonal design and the contamination rate and browning rate of every combination were calculated after the bulbets were cultured for 10 d. [ Result] The 4 selected factors all had very significant influence on the contamination rate ( P 〈 0.01 ) and among them, the bath temperature, surface disinfection time and kinds of surface disinfectant had significant influence on the contamination rate (P 〈 0.05 ). From the signal factor, the contamination rate was lower when the bath temperature was 40 ℃, and when the bulbets were treated by 0.1% mercuric chloride, or treated without preservative. The browning rate was lower when the bath temperature was 45 ~C and it could get better effect of decreasing the browning rate when the sur- face disinfection time of the treatment was short. [ Conclusion] The contamination rate was lowest under the condition of the bath temperature of 40 ℃, treating the bulbets by 0.1% mercuric chloride treating for 20 min and the medium without preservative.
