摘要
对引起花椒试管苗污染的外植体自身因素及生产过程中的环节进行了初步研究。结果表明 ,外植体在 4~ 5月份接种污染率最低 ,继代 2 0次 ,培养基中出现少量内生菌。培养环境对试管苗的污染有直接影响 (如人为活动、培养室的消毒、季节因素 ) ,试管苗在夏季污染率最高 ;封口材料相同时 ,三角瓶 (2 5 0ml)内试管苗的污染率 <塑料瓶 (330ml) <塑料瓶 (5 80ml) ;封口材料使用次数越多 ,污染几率越大 ;试管苗开口炼苗时间越长 ,污染率越高。
The elements causing contamination of Zanthoxylum bungeanum in virto culture were examined. The results were as follows : ( 1 ) the favorite seasons for implanting the explants with lowest contamination rate are April and May, in which there are a little endophytic bacterium after subcultured 20 times. (2)the culture environmental factors(for example people's activities, sterilization of the culture chamber and seasons) have the direct effect on contamination and the seedlings in test tube have highest contamination rate in summer. (3)the contamination rate is low in concial flask with the same seal materials (330ml plastic bottle 〈 580ml plastic bottle). (4)the higher contamination probability was, the more times the seal materials are used and the longer of the tissue to be open for exercise.
