松材线虫鉴定方法的研究与比较 被引量:6

摘要 通过形态学观察,同工酶分析,ITS-RFLP以及RAPD分析等方法分别对来自于不同国家和地区的40个线虫群体进行鉴定,并对四种分析方法进行比较.形态学观察可以初步区分松材线虫和拟松材线虫;通过谷草转氨酶的同工酶分析,可以准确地区分上述两种线虫;基于分子生物学的ITS-RFLP分析除了可以准确地鉴定两种线虫外,还可以对拟松材线虫的地域特征作出判断,划分为亚洲型和欧洲型;而RAPD分析不仅能准确地鉴定两种线虫,还可以描述各种松材线虫和拟松材线虫种内以及种间的遗传进化关系. The 40 isolates of pine wood nematodes were collected from various countries and areas, and identified by morphological characteristics, iso-enzyme analysis, ITS-RFLP and RAPD method respectively. Comparing with described above method, morphological characteristics could distinguish Bursaphelenchus xylophilus from Bursaphelenchus rnucronatus preliminarily iso-enzyme analysis of GOT was able to divide by rule and line two kinds of nematodes based on molecule biotechnology, ITS-RFLP method performed well in identifying two kinds of nematodes, and also in making judgments of terrain character of B. mucronatus which could be classified into Asian type or European type besides identified of B. xylophilus and B. mucronatus clearly, the RAPE) method was capable to describe the genetic relationship of interspecific and intraspecific in the evolution of each nematodes nicely.
