摘要
利用RAPD技术对松材线虫(Bursaphelenchusxylophilus)和拟松材线虫(B.mucrona-tus)的分离物基因组DNA进行了DNA片段的随机扩增,以确定种间及种内群体的亲缘关系和筛选具有种鉴定特征的分子标记.通过对40个10聚体随机引物的筛选,应用8个引物对两种线虫种间及种内分离物扩增片段的遗传分析显示:两种间群体RAPD扩增片段的共享度为40%~56%,而在松材线虫的不同分离物之间的共享度为60%~84%;其中有一个引物(P253)可在松材线虫和拟松材线虫种间产生具有种鉴定特征的特异性RAPD片段,在松材线虫的不同分离物中均可产生一约700bp的特异性片段,而在拟松材线虫中产生了两个分别约为1300bp和1900bp的特异性片段.
Two species in genera of Bursaphelenchus , B. xylophilus (Bx) and B. mucropatus (Bm), were examined for genetic relatedness among inter and infra species and screened for molecular marker with identical characteristics by random amplified polymorphic DNA (RAPD). Fourty ten mer random primers were screened and eight of which were used for genetic analysis. It is indicated that the similarity between inter species of Bx and Bm were 40% ̄56%, and the similarity between isolates of infra species of Bx were 46%~84%. A tested primer (P253) showed discriminating RAPD fragments between Bx and Bm isolates, i.e. a fragments (ca. 700 bp) for all the three Bx isolates and two fragments (ca. 1 300 bp and 1 900 bp) for Bm isolates.
