血管紧张素Ⅱ及其受体1在结直肠腺癌中表达及意义

The expression and significance of angiotensin-Ⅱ and angiotensin-Ⅱ type 1 receptor in colorectal carcinoma

摘要 目的探讨血管紧张素Ⅱ(angiotensin-Ⅱ,Ang-Ⅱ)及其1型受体(angiotensin Ⅱ type 1 receptor,AT1R)在人结直肠癌中的表达情况以及与Dukes分期的关系。方法收集2006年3月至2008年5月经手术切除的结直肠腺癌组织标本49例作为研究组,其中19例新鲜标本取其癌旁组织及肠断端正常组织为对照组1与2;将所有病例进行Dukes分期;用免疫组织化学染色观察Ang-Ⅱ、AT1R的分布;用RT-PCR检测AT1R mRNA表达。结果Ang-Ⅱ、AT1R蛋白在结直肠癌组织中的表达明显高于癌旁组织及正常组织(P均<0.01);AT1R mRNA与AT1R蛋白的表达具有明显正相关(r=0.534,P<0.01);AT1R蛋白的表达与Dukes分期无明显相关性(r=0.155,P=0.783)。结论Ang-Ⅱ、AT1R蛋白在结直肠腺癌组织中高表达,但其表达与肿瘤浸润的深度及淋巴结转移关系不大。AT1R可能可以作为结直肠腺癌基因治疗的一个新靶点。 Objective To explore the expression of angiotensin- Ⅱ ( Ang- Ⅱ ) and angiotensin- Ⅱ type 1 receptor ( AT1 R) in human colorectal carcinoma and analyze its correlation with Dukes stage of cnlorectal carcinoma. Methods Forty-nine colorectal carcinoma tissues,19 adjacent colorectal tissues and 19 normal colorectal tissues from 49 cases of colorectal carcinoma undergone operation from Mar. 2006 to May. 2008 were collected in this study. Dukes stages were carried out in all cases. The distribution of Ang- Ⅱ and AT1R was observed by immunohistochemistry (IHC) and the expression of ATt mRNA was observed by RT-PCR. Results Ang- Ⅱ and AT1R were mainly localized in cytoplasm or on cell membrane of glands cell. Compared with the normal and adjacent colorectal tissues, the Ang- Ⅱ and AT1 R positive cells in colorectal carcinoma tissues were significantly increased ( P 〈 0. 01 ) as well as the mRNA expression of AT1R was increased ( P 〈 0. 01 ). AT1R protein expression correlated with its RNA expression ,but its expression had no correlation with Dukes stage of colorectal carcinoma( r = 0. 155, P = 0. 783 ). Conclusions The expressions of Ang- Ⅱ and AT1R are high in colorectal carcinoma tissues, while have no correlation with tumor invasian depth or lymph metastasis.
