三七总苷对佐剂性关节炎大鼠腹腔巨噬细胞释放一氧化氮的影响 被引量:1

摘要 [目的]研究三七总苷(PNS)对佐剂性关节炎(AA)大鼠腹腔巨噬细胞释放一氧化氮(NO)的影响。[方法]大鼠右后足跖皮内注射弗氏完全佐剂诱发大鼠AA模型,采用硝酸还原酶法检测AA大鼠腹腔巨噬细胞释放的NO。[结果]体外给药PNS0.4mg/ml,作用4h时,增加AA大鼠NO释放(P<0.05);体外给药PNS 0.2mg/ml及0.4mg/ml,作用8h时,抑制AA大鼠NO释放(P<0.05,P<0.01);体外给药PNS 0.8mg/ml及PNS 1.6mg/ml,作用24h时,增加AA大鼠NO释放(P<0.01,P<0.05)。[结论]PNS体外给药随浓度及作用的时间变化,对AA大鼠释放NO有双向调节作用,PNS抑制AA大鼠腹腔巨噬细胞释放NO可能在AA治疗中起一定的作用。 [Objective]To investigate the influence of panax notoginseng saponins (PNS)on production of Nitric Oxide(NO) by peritoneal maerophages of rats with adjuvant arthritis(AA). [Methods]Complete Freund's adjuvant was used to induce AA in rats. Pro duetion of NO by peritoneal maerophages of rats with adjuvant arthritis was determined by nitrate reductase. [Results]PNS 0.4mg/ ml increased NO synthesis and secretion from peritoneal macrophages of AA rats in vitro after four hours(P〈0.05) ;PNS 0.2mg/ ml and 0.4mg/ml reduced NO synthesis and secretion from AA rats in vitro after eight hours respectively(P〈0.05, P〈0.01); PNS 0.8mg/ml and 1.6mg/ml increased NO synthesis and secretion from AA rats in vitro after twenty-four hours respectively(P〈0.01 ,P〈0.05). [Conelusion] PNS has bidireetion influence on production of NO by peritoneal macrophages of AA rats with change of concentration and times. PNS reduced NO synthesis and secretion from peritoneal maerophages of AA rats, which may be effective on treating AA.
