摘要
利用黄酮类化合物可与金属离子形成有色螯合物的原理,建立果蔬纤维颗粒中总黄酮含量的测定方法并进行样品制备最佳条件选择及测定。正交设计选择最佳样品制备条件,采用分光光度法,KAc-AlCl3体系与样品中黄酮提取物反应,选择最大吸收波长,以芦丁为对照品制作标准曲线,并作方法学考察。品制备选择70%乙醇溶液,提取温度80℃,回流3h,用KAc-AlCl3体系比色法测定果蔬纤维颗粒中总黄酮含量为0.89%,精密度RSD=0.77%(n=5),平均回收率为101.04%,RSD=1.21%(n=5)。结论:KAc-AlCl3体系比色法测定果蔬纤维颗粒总黄酮含量方法可行,呈色稳定,操作简便,适用于生产过程产品质量监控。
To establish the methods of determination of total tlavones in Guoshu fiber granules and choose the optima conditions on sample preparation and determination according to colorful chelate formed by flavone and metal ion. ptima conditions of sample preparation were determined by mating design; spectrophotometry was conducted; KAC-AlCl3 system was reacted with flavones extract from samples; the maximum wavelength was selected; standard curve was made with Rutin---the control article; and methodology was investigated, sample preparation was under the condition of 70 % ethanol, 80℃, and recirculation 3 h; the content of total flavones was 0.89 % in Guoshu fiber granules via KAC-AlCl3 system colorimetry and RSD was 0.77 % (n=5); the average recovery rate was 101.04 % and RSD was 1.21% (n=5). Conclusion: the method of determination of total flavone in Guoshu fiber granules via KAC-AlCl3 system colorimetry was feasible with stable color and simple conduction, which is suitable for product quality monitoring during process of production.
