摘要 目的:确定内蒙古JSRV-NM病毒株在逆转录病毒与宿主共进化史上的地位。方法:从我国内蒙古羊种绵羊肺腺瘤病羊肺组织标本中分离出exJSRV-NM株。PCR方法扩增出exJSRV-NM和enJSRV-NM株全长cDNA并进行全基因组测序。应用DNAstar软件对JSRV-NM基因组上的U3、ENV、GAG/POL核苷酸序列进行分析。结果:克隆的exJSRV-NM株和enJSRV-NM株全基因全长分别为7462bp和7430bp。序列分析表明,exJSRV-NM株序列与Gen-Bank上收录的南非代表株基因组和JSRV21核苷酸全序相比其同源性分别为93.3%和99.2%。enJSRV-NM株序列与GenBank上收录的enJS56A1相比其同源性为94.8%。结论:我国内蒙古羊种所携带的JSRV-NM在逆转录病毒与宿主共进化史上属于古老型。 Objective: To identify the evolving place of jaagsiekte retrovirus-NM(JSRV-NM) in the retrovirus-host co-evolution history based on the sequencing analysis of exogenous JSRV-NM(ex JSRV-NM) and endogenous jaagsiekte retro-virus-NM(en JSRV-NM). Methods: The tissue samples of sheep pulmonary adenocarcinoma were collected in Inner Mongolia. The exJSRV-NM was isolated form the tissue samples. The full length cDNA of exJSRV-NM and enJSRV-NM was amplified by PCR using several pairs of primers designed according to the published references. The PCR products was sequenced. The nucleotide sequences of GAG/POL gene,ENV gene,and U3 gene were compared with those of 21 JSRV strains published on the GenBank using DNAstar software. Results: The exJSRV-NM and enJSRV-NM were cloned and sequenced successfully. And their full length were 7 462 bp and 7 430 bp respectively. The nucleotide homology was 93.3% between the exJSRV-NM and South Africa strain,99.2% between exJSRV-NM and JSRV21. And the nucleotide homology was 94.8% between enJSRV-NM and enJS56A1. Conclusion: The JSRV-NM isolated form Inner Mongolia sheep of our country belongs to the ancient type in sheep evolving history.
