摘要
方法:成年SD大鼠24只,分为3组,每组8只,分为安静对照组、运动对照组、运动加药组,建立运动应激性胃溃疡动物模型。对各组大鼠胃黏膜组织进行病理损伤指数的计分,经免疫组化染色分析胃黏膜组织中HSP70表达变化;制备胃黏膜组织匀浆,检测超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果:与安静组相比,运动对照组大鼠SOD活性明显下降,MDA含量显著增加,HSP70表达稍有增加;加药组大鼠SOD活性和MDA含量变化不显著,HSP70表达显著增加;与运动对照组比较,加药组SOD活性明显上升;MDA含量明显下降;HSP70表达进一步增加。结论:白术可以预防运动应激性胃溃疡,其保护机制可能与降低大鼠胃组织中自由基含量,增强SOD活性和提高HSP70的表达有关。
Objective: To investigate the exercise stress induced ulcer in the process of free radical change and content of the expression of HSP70 and the influence of Rhizoma Atractylodis Macrocephalae on the prevention and treatment of exercise induced gastric ulcer and its mechanism. Method: 24 adult SD rats, divided into three groups, quiet control, movement control, and exercise plus medicine group, each group has 8. And established the exercise induced ulcer model. The rats in each group were calculated the ulcer index and inhibition of ulcers, the SP stained by light microscopy study of organizations; respectively hydroxylamine law and sulfur barbiturates reaction detection of gastric Organization superoxide dismutase activity and the MDA content, immunohistochemistry method measured gastric tissue in the heat shock proteins (HSP) 70 expression. Results: Compared with the quiet, movement control rats SOD activity decreased significantly, MDA was significantly increased, HSP70 expression of a slight increase in rats and SOD activity decline was not obvious, MDA content does not increase significantly, HSP70 expression increased significantly; exercise control group, adding medicine group SOD activity was significantly increased; MDA were significantly decreased;HSP70 expression increase. Conclusion: Rhizoma Atractylodis Macrocephalae can prevent the stress ulcer and reduce the role of free radicals in the stomach contents, enhance the activity of SOD and HSP70 expression.
