摘要
目的:观察不同浓度五倍子水提取物对内毒素诱导人牙髓细胞分泌IL-6的影响。方法:采用组织块法体外培养人牙髓细胞,以含不同浓度五倍子水提取物(1.25、2.5、5、10、20μg/mL)、25μg/mL内毒素和20mL/L新生牛血清的DMEM培养基作用于第5代人牙髓细胞,用放射免疫法测定IL-6含量,采用单因素方差分析(完全随机设计)和t检验对实验数据进行统计学分析。结果:低浓度(1.25、2.5、5、10、20μg/mL)五倍子水提取物能明显抑制25μg/mL内毒素诱导人牙髓细胞分泌IL-6,这种抑制作用在一定范围内呈浓度依赖性。结论:低浓度五倍子水提取物具有抑制内毒素诱导人牙髓细胞分泌IL-6的作用。
AIM: To observe the influence of Chinese nutgall extracts in different doses on the effect of lipopolysaccharide upon the secretion of IL -6 from human dental pulp cells. METHODS: Human dental pulp cells were cultivated with tissue explant. Human dental pulp cells of the fifth passage were cultured in DMEM with 25 μg/ml lipopolysaccharide, Chinese nutgall extracts( 1.25,2.5,5,10, 20 μg/ml) and 20ml/L new born bovine serum. Radioimmunoassay (RIA) was applied to determine the amount of cells' IL- 6. One -way ANOVA( completely random design) and t - test were used in statistical analysis. RESULTS : Low - dose of Chinese nutgall extracts ( 1.25,2.5,5, 10,20 μg/ml)significantly inhibited the effect of lipopolysaccharide on the human dental pulp cells'secretion of IL -6 in a dose - dependent manner. CONCLUSION: Low - dose Chinese nutgall extracts can inhibit the effect of lipopolysaccharide on the secretion of IL -6 from human dental pulp cells .
