摘要
目的:采用原核表达系统表达抗菌肽葛佬素,并观察其对原代牙髓细胞炎性细胞因子IL-1β分泌的影响。方法:应用pET原核表达系统进行目的蛋白的表达,应用蛋白免疫印迹对蛋白进行鉴定,并采用ELISA方法,观察重组葛佬素对内毒素诱导的牙髓细胞IL-1β释放的影响。结果:原核表达产物经蛋白免疫印迹鉴定,为融合有聚组氨酸标签的葛佬素抗菌肽,ELISA检测结果表明,该表达产物能够以剂量依赖方式抑制LPS诱导的IL-1β释放。结论:在原核表达系统中成功表达了具有生物活性的抗菌肽葛佬素,该表达产物可以抑制牙髓细胞炎性因子IL-1β的释放。
AIM:To express anti-bacterial peptide gloverin in prokaryotic system and investigate its anti-LPS effects in pulp cells.METHODS:E.coli BL21 (DE3) was used as to express gloverin,Western blot was used to identify the production and ELISA was used to detect the effects of gloverin on IL-1β release in pulp cells.RESULTS:The production was identified as gloverin peptide by Western blot,and the ELISA data indicated that the recombinant gloverin could inhibit LPS induced IL-1β release in pulp cells.CONCLUSION:Anti-bacterial peptide gloverin was successfully expressed in E.coli BL21,the recombinant polypeptide has anti-LPS effects in pulp cells.
