C/EBPβ基因与PPARγ基因在胎猪脂肪组织和原代培养的脂肪细胞中的表达 被引量:2

摘要 研究采用实时定量RT-PCR方法对发育到30,60,90日龄的胎猪脂肪组织中C/EBPβ与PPARγ基因的表达情况进行检测,并且对2个基因在原代培养的脂肪细胞中的表达情况进行检测。结果显示:在30,60日龄胎猪的脂肪组织中并没有检测到C/EBPβ基因的表达,在90日龄胎猪的脂肪组织检测到其低水平表达;PPARγ基因在30日龄胎猪的脂肪组织中已经表达,并且在60,90日龄胎猪的脂肪组织中仍然持续表达。在原代培养的前脂肪细胞中,2个基因均表达;诱导后C/EBPβ基因的表达量在24小时达到高峰,48小时和72小时时表达量显著回落;诱导后PPARγ基因的表达量明显升高,24小时达到高峰,之后随着培养时间的延长表达量逐渐降低。 The expression of C/EBPβ and PPARγ gene at 30,60 and 90 days in pig fetal tissue and primary stromal - vascular cultures of Yorkshire were analyzed by real - time quantitative RT - PCR. The expression of C/EBPβ gene was not detected at 30 and 60 days fetal adipose tissues and was detected only at low levels in 90 days fetal adipose tissues. The expression of PPARγ gene was detected as early as 30 days of fetal development in adipose tissue and continued through 90 days. Both of C/EBPβ and PPARγ gene were detected in primary stromal - vascular cultures, the expression of C/EBPβ gene was get peaked around 24 h postinduction, and it was quickly diminished after this ; the expression of PPARγ gene was up - regulated gradually after cultured and then obviously increased till 24 h postinduction, and the expression of PPARγ gene was down - regulated gradually with culturing continued
