Determination of Free Amino Acid Content in Different Litchi(Litchichinensis Sonn.) Varieties Farina by RP-HPLC Method

摘要 [目的]通过研究荔枝花粉中的氨基酸含量,促进对荔枝花粉资源的开发利用和了解影响不同荔枝品种品质形成的因素。[方法]采用柱前衍生RP-HPLC测定4个荔枝品种(桂味、怀枝、黑叶、妃子笑)花粉的全谱游离氨基酸,以邻苯二甲醛(OPA)与9-芴基羰酰氯(FMOC—CI)作衍生剂,正缬氨酸为内标,利用二极管阵列(DAD)可变波长检测程序。[结果]各种氨基酸的保留时间(Rt)RSD为0.01%。3.02%,相对峰面积(Ax/AI)RSD为1.19%~7.82%,回收率为95.21%~114.24%。4种荔枝品种花粉的总游离氨基酸含量从大到小排序为(mg/100mg):桂味26.88、淮枝21.46、黑叶20.92、妃子笑18.91。[结论]总游离氨基酸含量的不同会影响到荔枝品质,其中桂味总游离氨基酸含量最高,营养价值高,可在养蜂业和花粉营养品加工业中加以充分利用。 [ Objective] The research aimed to study the content of free amino acid in different litchi ( Litchichinensis Sonn. ) varieties farina, promoting the development and utilization of Litchichinensis Sonn. farina resources and knowing the quality of different varieties of litchi form factor. [Method] The whole free amino acids in farina of 4 Litchichinensis Sonn. varieties were assayed by using RP-HPLC method. In this method, Ortho-phthalaldehyde(OPA) and 9-fluorenylmetloyl chloroformate( FMOC ) were selected as pre-column derivative reagent, Norvaline as internal standard, and diode army (DAD) variable-wavelength as measure function. [Result] The results showed that the RSD for Rt and Ax/AI of amino acids was from 0.01% to 3.02% and from 1.19% to 7.82% respectively. In the present stud)', the recovery ratio was from 95.21% to 114.24%. Among 4 Litchi.farina varieties, the content of total free amino acids from high to low was Guiwei 26.88 mg/100mg, Huizhi 21.46 mg/100 mg, Heiye 20.92 mg/100 rag, and Feizixiao 18.91 mg/100 mg. [Conclusion] The free amino acid content of different quality would affect Litchichinensis Sonn.. Thereinto, the flee amino acid of Guiwei was the highest, had high nutritional value. So peple could take full advantage of apicuhure and pollen nutrition industry.
