摘要 为了研究甘薯悬浮培养细胞黄酮类物质的积累,利用甘薯块根诱导愈伤组织,在黑暗条件下建立悬浮细胞培养体系,通过测定细胞密度和黄酮类物质含量来确定甘薯悬浮细胞的培养条件.结果表明,甘薯悬浮细胞黄酮类物质的含量同甘薯的块根(0.69%,干重)相当,各种培养条件下的平均值为0.64%;理论最佳培养条件为:用添加2.0 mg·L-12,4-D的固体MS培养基诱导愈伤组织,经多次继代培养后,在接种量为2 g?20 mL-1、2,4-D为2.0 mg·L-1的液体培养基中进行摇悬浮培养,其悬浮细胞黄酮类物质的含量达到0.79%. In order to comprehend flavonoid accumulation of suspension culture cells of sweetpotato, the calluses were induced by using the tuberous roots of sweetpotato, and the suspension cell system was erected from the calluses under dark condition. For acquired suspension cells, the condition of suspension culture was screened by determining cell density and flavonoid content. The results indicate that the flavonoid content of suspension cells was consistent with that of the tuberous root of sweetpotato (0.69%, dry weight), mean of flavonoid of suspension cells was 0.64% under all culture conditions. Optimized conditions were theoretically obtained too: the calluses were induced by MS solid culture medium containing 2.0 mg·L^-1 2, 4-D during subcultures, and then 2 g · 20 mL-1 calluses were inoculated to MS liquid culture medium containing 2.0 mg·L^-1 2, 4-D, which flavonoid content of suspension cells was 0.79%,
