摘要
目的观察鹿茸方对糖尿病肾病大鼠肾皮质转化生长因子-β1(TGF-β1)基因表达的影响,在基因水平探讨鹿茸方疗效的分子机制。方法应用雄性Wistar大鼠腹腔注射链脲霉素诱导糖尿病肾病动物模型,造模成功后分为模型组、鹿茸方小剂量治疗组、鹿茸方中剂量治疗组、水飞蓟宾治疗组及正常对照组。干预治疗8周后,测定各组大鼠24h尿微量白蛋白排泄量(UAER)、用酶联免疫吸附法测定各组大鼠肾皮质TGF-β1;用RT-PCR法测定各组大鼠TGF-β1mRNA的表达。结果模型组大鼠UAER、肾皮质TGF-β1mRNA及蛋白表达显著增高,均较正常大鼠显著上调(P<0.05);鹿茸方中、小剂量组显著降低模型UAER、肾皮质TGF-β1含量,显著下调肾皮质TGF-β1mRNA的表达(P<0.05)。结论鹿茸方及对照药物水飞蓟宾均对糖尿病肾病有一定治疗作用,其作用机制与下调肾皮质TGF-β1mRNA的表达有关。
Objective To study the effect of Lurong decoction on the mRNA expressions of TGF- β1 in renal cortex of diabetic rats and to explore the molecular mechanism of Lurong decoction for treatment of diabetic nephropathy, Methods Forty male wistar rats were intraperitoneally injected with streptozotocin (5 mg/kg) to induce diabetic models, These models were divided into 4 disposal groups randomly,. Pathologic group, low dose group, middle dose group, silibinin group. Five rats were chosen as normal group, After treatment of eight weeks, experimental rats were killed. Urinary albumin excretion rate (UAER) and renal cortex transforming growth factor - betal were measured, The mRNA expressions of transforming growth factor - betal (TGF - β1 ) were assessed by RT - PCR analysis. Results Compared with normal group, the levels of UAER, renal cortex TGF - β1 as well as mRNA expressions of TGF - β1 increased significantly in pathologic group (P〈0.05). Compared with pathologic group, UAER, renal cortex TGF - β1 and mRNA expressions of TGF- β1 were downregulated significantly in treatment group (P〈0.05), Conclusion Lurong decoction and silibinin had the preventive effect on diabetic nephropathy, The effects were associaled with downregulating the mRNA expressions of TGF- β1
