摘要
注射外源FSH诱导羔山羊卵泡发育并活体采卵,比较不同来源FSH、不同采卵间隔时间、不同重复采卵次间的实验效果。结果显示:1)用5 mg新西兰产FSH(Ovagen)处理后每只羊卵巢上平均有(18.3±3.5)个可用卵泡,较中科院产FSH效果(14.9±6.7)好。2)间隔7 d或14 d进行卵泡诱导发育及活体采卵,实验效果无显著性差异。3)羔山羊共4次用5 mg FSH(Ovagen)重复诱导卵泡发育并活体采卵,随次数增加,可用卵泡数和回收可用卵母细胞数呈下降趋势;不同重复次间卵泡直径、回收卵母细胞的可用率无显著性差异(P>0.05)。4)间隔7 d多次重复采卵过程中,平均每次采卵获可用卵8.4枚,COCs回收率和可用率分别为74.2%、78.5%;不同个体间卵母细胞回收率和可用率无显著性差异(P>0.05)。结论:新西兰产FSH(Ovagen)比中科院产FSH诱导羔山羊卵泡发育效果好;手术法活体采卵可获得稳定的卵母细胞回收率和可用率,羔山羊可以进行手术法重复活体采卵且最短采卵间隔时间宜为7 d。
This study was performed to examine the follicular development of kids aged 120 to 150 days stimulated with different follicule stimulating hormone (FSH) products and the optimal regime of laparotomy oocytes recovery from live donors. The result showed that the number of useable follicles (diameter〉2 mm) was 18.3± 3.5 per kid when kids were stimulated with 5mg FSH (Ovagen, ICP, New Zenland) and 14. 9± 6.7 in kids treated with 5 mg FSH (Chinese Academy of Science, Beijing). There was a significant difference between the two groups (P〈0.05). The interval of ovum pick-up did not affect its efficiency. There were no significant difference in the number of useable follicles ((13.0 ± 4.2) vs (13.4± 4.3)), average diameter of follicle ((4.68 ±1.46) mm vs (4.92 ± 1.63) mm), recovery rate of oocytes ((78. 1 ± 14. 9) % vs (77.0±9.9)%), and useable rate of oocytes ((78.4±12.0) % vs (75.6 ±15.6)%) between the OPU intervals at 7 d and 14 d. Eight kids aged 131 to 150 days were administrated with 5mg FSH (Ovagen) and ovum pick-up repeatedly. The usable follicles (diameter 〉2 mm) were 20. 1 ± 5.6 in OPU1, 15.5± 3.2 in OPU2, 9.3±3.7 in OPU3 and 8.7±2.5 in OPU4 (P〈0.05), while the usable oocytes were 11. 1 ±3.2, 8.3±1.8, 6.6± 3.7 and 6.0 ± 1.7 (P〈0.05), respectively. The number of usable follicles from OPU per kid decreased with increasing OPU times. There was no significant difference in the follicular diameter and usable rates in each OPU. A total of 279 oocytes were recovered from 376 follicles and about 219 of them could be useful for in vitro maturation in 26 OPUs of eight kids. About 8.4 usable oocytes were recovered per OPU. Both rates of recovery and usable oocytes were 74.2% and 78.5% respectively. No significant differences were observed in recovery and usable rate among donors.
