摘要 为提高生物素缺陷型菌株发酵生产谷氨酸的单罐产量,对发酵罐搅拌器和高生物素谷氨酸发酵工艺进行研究。试验结果表明:将100m3发酵罐原有6弯叶圆盘涡轮搅拌器改造为6半圆叶圆盘涡轮搅拌器,并对其尺寸进行优化,发酵罐的溶氧系数提高了147%;通过在最终改造的发酵罐中进行发酵试验,其适宜的初始生物素浓度和单罐谷氨酸产量均达到最高,分别为12.2μg/L和11.65t,比搅拌器改造前分别提高了56%和25%。因此,提高发酵罐的溶氧系数,有利于提高生物素的适宜用量,最终促进了单罐谷氨酸产量的显著提高。 In order to increase the yield of glutamate in the single fermentor with biotin-auxotrophic bacteria, the stirrer of fermentor and glutamate fermentation process with high concentration biotin were investigated. The experiment results showed that the dissolved oxygen coefficient was increased 147% by taking six half round blade disk turbine stirrer instead of six tortile blade disk turbine stirrer in 100 m^3 fermentor, and the dimension of the stirrer were optimized. Moreover, by carrying out experiments in the improved fermentor, the fitting initial concentration of biotin and the yield glutamate in single fermentor were maximized, up to 12.2 μg/L and 11.65 t, which were increased 56% and 25% separately. As a resuit, increasing the dissolved oxgen coefficient can help to increase the fitting biotin concentration, and the yield of glutamate in single fermentor was increased obviously.
