菊花中D-甘露醇及糖类成分的毛细管电泳电化学检测研究 被引量:7

摘要 利用毛细管电泳-电化学检测研究菊花中的糖类,并首次测定出D-甘露醇的含量。比较了不同种菊花中D-甘露醇及6种糖含量的差异。研究了电极电位、运行液的浓度、电泳电压及进样时间等对D-甘露醇及糖类成分分离的影响,得到了较优化的测定条件。以直径0.300mm的铜圆盘电极为检测电极,工作电极电位0.65V(vs.SCE),0.1mol.L-1NaOH运行液中所列组分完全分离,检测限(S/N=3)2.5×10-7~0.8×10-6g.mL-1之间。野菊电泳图谱在15.5~20.0min之间具有独特且相对稳定的特征峰。 A high performance capillary electrophoresis (CE) combined with electrochemical detection (ED) has been employed to study and compare the content of saccharide and D-Mannitol firstly in chrysanthemum. The effects of some important factors such as the potential applied to the working electrode, the concentration of the running buffer, separation voltage and injection time are investigated to choose the CE-ED optimum conditions. Operated in a wall-jet configuration, a 300 μm diameter copper-disc electrode is used as the working electrode, 0.65 V(vs.SCE), and the running buffer is 0.1 mol·L^-1 NaOH. The detection limit (S/N=3) is between 2.5×10^-7~0.8×10^-6g·mL^-1. The electropherograms of Flos chrysanthemi Indicium collected from different regions in China have their characteristics and stable fingerprint area during 15.5 to 20 min.
