摘要
目的探讨高压氧(HBO)对缺氧缺血新生大鼠神经细胞凋亡及caspase-3表达的影响。方法建立缺氧缺血性脑损伤(HIBD)大鼠模型,将其分为HIBD组及HBO处理组,并设立假手术组(n=24)。观察3组大鼠不同时间脑组织病理学改变,同时用免疫组化的方法检测3组大鼠脑组织内caspase-3蛋白表达的动态变化。结果HIBD组18、24、48和96h时间点海马及皮层区caspase-3蛋白的表达均明显高于假手术组(P<0.05);HBO干预组大鼠caspase-3表达的升高程度均低于HIBD组(P<0.05)。结论HBO能降低大鼠海马及皮层区神经细胞caspase-3蛋白的表达水平。
Objective To explore the effect of hyperbaric oxygen (HBO) on the apoptosis resulted from hypoxicischemic brain damage (HIBD) and potential relation to caspase-3 expression. Methods Rats were divided into HIBD.HBO.sham operation group. To observe the morphologic change of brain tissues treated by HBO and compared with that of the hypoxia-ischemia rat model, then check the dynastic change of caspase-3 expression by immunohistochemistry at different time point. Results The expression of caspase-3 in the hippocarnpus and cortex was higher than that of sham group at 18, 24, 48 and 96 h time point( P 〈 0. 05 ). The increased caspase-3 expression decreased significantly in the brain of HIBD rats after HBO treatment (P 〈 0. 05). Conclusion HBO inhibited the expression of caspase-3 in hippocampus and cortex of HIBD rats.
