摘要
目的探讨大鼠角膜真菌感染后角膜局部细胞因子的表达。方法在24只大鼠左眼角膜上制作烟曲霉菌性角膜炎(AFK)模型为实验组,右眼为对照组。在感染后早、中、晚期取下角膜组织,采用逆转录聚合酶链式反应(RT-PCR),检测白细胞介素-6(IL-6)、白细胞介素-10(IL-10)在不同病程角膜组织中的表达。结果在实验组角膜中,IL-6早期即大量表达,中期炎症反应高峰时达高峰,以后逐渐下降;IL-10在早期表达最高以后逐渐下降。结论IL-6在AFK病程中是一个敏感的炎性因子,与局部的炎症反应程度一致。IL-10在AFK病程中呈逐渐下降趋势。
Objective Keratomycosis is a leading sight-threatening infection of the corneal blindness.Our previous research showed that immune and hypersensitivity reactions play a role in the progression of fungal keratitis.The aims of this study was to identify the kinetics of infiltrated cytokines in the aspergillus fumigatus keratitis.Methods A rat model of fungal keratitis was established by smearing the aspergillus fumigatus in the cornea of the left eyes in 24 Wistar rats,and the normal saline solution was used in the right eyes as control.The histological change and spore and hyphal of funga were examined on the 2nd,6th and 14th day after modeling under the microscope.Pro-inflammation cytokine-IL-6 and anti-inflammation cytokine-IL-10 were detected using RT-PCR in the different course.Results The infiltrated immunity cells and the fungi grew increasedly from the early stage and reached peak at the midterm stage.At the later stage,cornea leucoma and lots of neovascularization were seen,but no fungi was found.IL-6,IL-10 were weakly expressed on normal control group.However,large amount of IL-6 were detected in the 2nd day of aspergillus fumigatus keratitis,and the highest expression level was revealed at the 6th day with the severest inflammation reaction,showing a significant difference between various stage of aspergillus fumigatus keratitis and the control group（P〈0.01）and a intergroup significant difference（P〈0.01）.The peak expression of IL-10 was at the 2nd day and showed a gradual decline at the 14th day of aspergillus fumigatus keratitis.A statistically significant difference was seen between various stage of keratitis（P〈0.01）.Conclusion The overexpression of IL-6 in cornea with aspergillus fumigatus keratitis is parallel to the extent of inflammation reaction.The peak expression of IL-10 is at the early stage of aspergillus fumigatus keratitis.
