桃属植物等位酶遗传变异分析及品种基因型指纹利用 被引量:3

摘要 利用不连续双垂直板聚丙烯酰胺凝胶电泳技术,对国家种质郑州桃资源圃所收集到的97份桃(AmygdalusL.)材料和2份近缘种李(Prunus salicinaL.)共计99份材料进行了等位酶遗传变异分析。结果表明:①在11个酶系统中共检测到24个清晰位点和69个等位基因,多态位点为22个,位点最大等位基因数为8,体现出桃资源丰富的遗传多样性。②不同的分类群具有特有等位基因:如Est-2a、Est-2d、G6pdh-1a、Mdh-1c、Pod-3b、Pod-3d、Sod-3a、Sod-3b仅存在于普通桃中,Cat-1h、Est-2f、G6pdh-1b、Me-1a只存在于山桃中,Cod-3c、Est-3c、Me-1c、Me-2b只存在于光核桃,体现了桃种间的遗传组成差异。③利用11个酶系统的24个位点的69个等位基因所构建的桃品种(株系)等位酶基因型指纹可以将99份试材完全区分,表明等位酶是桃品种鉴定的一种有效工具。 Allozymic variation of 97 peach materials and 2 related species plum were., investigated using vertical slab poly-acrylamide gel electrophoresis. The results showed that:①Sixty-nine alleles of 24 loci in 11 enzyme systems were detec-ted. 22 loci are polymorphic. The maxium number of alleles in a locus was eight. The result demonstrated a rich geneticdiversity in peach germplasm.②some specific alleles were detected in different outgroup, such as Est-2^a, Est-2^d, G6pdh-1^a, Mdh-1^c, Pod-3^b, Pod-3^d, Sod-3^a, Sod-3^b were only found in Amygdalus persica L. and Cat-1^h, Est-2^f, G6pdh-1^b,Me-1^a only in Amygdalus davidiana (Carr.) Y , while Cod-3^c, Est-3^c, Me-1^c, Me-2^b occurred in Amygdalusrnira Koe-hne. The result also indicated that genetic composition was highly differentiated among different species in genus Amyg-dalus L.③Based on multi-locos genotype fingerprinting cuhivars with 69 alleles at 24 loci, 99 of materials were clear i-dentified, suggesting allozyme was a useful marker for peach cuhivars identification.
