AM真菌孢子萌发及其与白三叶草双重培养研究初报 被引量:5

摘要 以4种AM真菌繁殖体为培养对象,通过不同低温处理,对孢子萌发率进行了比较,并对Gigas-pora margarita孢子与白三叶草离体根系双重培养也进行了试验,结果表明:(1)土著孢子在4℃贮藏10~50d均显著提高了孢子萌发率,处理20d萌发效果最佳,萌发率为30.34%;(2)4种AM真菌的繁殖体形式对孢子萌发的影响存在显著差异,萌发率高低次序为Gl.sp孢子果>Gi.margarita>Gl mosseae>Gl.sp孢子;Gigaspora margarita孢子与白三叶草离体根系双重培养系统中,根系对孢子萌发影响不显著,但明显促进了菌丝的分支,并观察到菌丝对根系典型的匍匐侵染现象;停止生长的厚壁菌丝下方产生有隔分支菌丝,逐步发展为分支吸收结构(BAS)。培养1个月后被侵染的根系产孢率为(45.76±5)个/毫米根长。菌丝的生长方向不受根系的影响,即厚壁菌丝具有负向地性,分支菌丝为向地性。 The germination ratio of spores was compared after stored different time in cool temperature and with different progenitive form.The dual culture of Gigaspora margarita spores and clover roots was done as well.The results showed:(1) The native spores stored at 4℃ for 10-50 days significantly gained higher germination ratio.The highest ratio was 30.34% in the track of 20 days store.(2) Different progenitive form had different germination ratio.In this experiment,the order was Gl.sp sporacarps〉Gi.margarita〉Gl.mosseae〉Gl.sp spores;(3) In dual culture,the effect of host plant roots on spore’s germination was not significant,but obviously promoted branch of hypha.The groveling infection was observed.Under runner hypha which have stopped growing branched separated ones,and further developed branched absorbing structure.After one month,there were(45.76±5) spores per millimeter root.The hypha growth direction was not influenced by roots.
