摘要 目的研究创伤后应激障碍(PTSD)大鼠下丘脑内促肾上腺皮质激素释放因子(CRF)和精氨酸加压素(AVP)表达的变化。方法采用国际认定的SPS方法刺激大鼠,建立PTSD大鼠模型,分别取SPS处理后24h,7d,14d大鼠下丘脑组织;同时取正常下丘脑组织作为对照,应用免疫组化、Westernblot方法分别观察各组下丘脑神经元CRF和AVP表达变化。结果PTSD大鼠下丘脑神经元CRF表达随着时间推移逐渐增加,而后表现出恢复趋势,于14d时趋于正常。AVP具有与CRF相同的变化趋势。结论PTSD大鼠下丘脑CRF和AVP表达上调,提示CRF在AVP的协同下调节糖皮质激素水平变化,可能影响PTSD精神症状的表现。PTSD大鼠下丘脑内CRF和AVP的表达变化可能与海马神经元盐皮质激素受体(MR)和糖皮质激素受体(GR)变化相关。 Objective To invest the expression of CRF and AVP in the hypothalamic neurons in PTSD rat. Methods PTSD rat model was set up by stimulating the rat with SPS-method confirmed by international medical organization. The hypothalamus tissues of rats were taken on 24 h, 7 d, 14 d after treated with SPS and the hypothalamus tissues of normal rat brains were taken as the control group. With the and western blotting technique, the expression changes of CRF and AVP in the hypothalamic neurons in each group were observed and detected. Results After treated with SPS, the expression of CRF in hypothalamic neurons gradually increased on 24 h and 7 d, and then returned to normal on 14 d. AVP had the same effect as CRF. Conclusion Treated with the SPS, the expressions of CRF and AVP are up-regulated in hypothalamus of rots, which reveals that CRF regulates glucocorticoid levels with the synergy of AVP. They may affect the Psychiatric symptoms of PTSD. And in PTSD rats' hypothalamus, the expressions of CRF and AVP may be related to hippocampal MR and GR.
