摘要
目的探讨沙利度胺(Thd)对类风湿关节炎(RA)患者外周血T淋巴细胞的免疫调节作用。方法在植物血凝素(PHA)刺激下,RA患者外周血T淋巴细胞与不同浓度的Thd(2.5、25、100、300、500μg/ml)共同培养。用四甲基噻唑蓝(MTT)比色法检测T淋巴细胞的增殖,用流式细胞术检测T淋巴细胞的凋亡和T淋巴细胞CDl52、CD28的表达情况,用Real—time聚合酶链反应(PCR)检测T淋巴细胞的白细胞介素(IL)-6、IL-10、肿瘤坏死因子(TNF)一QmRNA表达水平。结果在体外,与阴性对照组相比,500μg/ml Thd组显著抑制T淋巴细胞的增殖,促进T淋巴细胞早期凋亡,抑制T淋巴细胞CD3+CD28+表达,促进CD8+CD152*3K达,而≤300μg/ml,对T淋巴细胞未见明显影响。与对照组相比,(100~500)μg/ml剂量组可抑制T淋巴细胞IL-6、TNF—dmRNA表达,(2.5~500)μg/ml可促进IL-10mRNA的表达。结论Thd可能通过影响RA患者外周血T淋巴细胞增殖、早期凋亡、CD3+CD28+和CD8+CD152+表达,以及IL-6、IL-10、TNF—dmRNA表达,反转RA患者的免疫平衡失控。
Objective To study the immunoregulatory effects of thalidomide on the peripheral blood T-lymphocytes of rheumatoid arthritis patients. Methods MTT was used to detect the effects of different thalidomide concentrations on the proliferation of T-cells. Flow cytometry was used to analyze T-cells early apoptosis and the T-cells subsets in different concentration of thalidomide. The mRNA expression of IL-6,1L- 10 and TNF-α was measured by RT-PCR method. Results The level of thalidomide at 500μg/ml inhibited the proliferation of T-cells and the CD3+CD28+ expression of T-cell subsets, but promoted the early apoptosis and the CD8+CD152+ expression of T-cell subsets. Thalidomine at any concentration could inhibit the mRNA expression of IL-6,TNF-α. However, the level of thalidomide that could promote the mRNA expression of IL- 10 was 100 μg/ml and 500 μg/ml, Conclusion Thalidomide can inhibit the proliferation of T lymphocytes and the expression of CD3+CD28+ on T-cell subsets. It can promote the early apoptosis and the CD8+CD152+ expression of T-cell subsets. Thalidomide inhibits the mRNA expression of IL-6 and TNF-α but promote the mRNA expression of IL-10. Thalidomide has immuno-regulatory effects on rheumatoid arthritis T-cells.
