摘要
[目的]建立HPLC测定盐酸多西环素片的含量。[方法]采用ODS柱(250mm×4.6mm,5μm),以0.05mol/L草酸铵溶液︰二甲基甲酰胺︰0.2mol/L磷酸氢二铵溶液(65︰30︰5)(用氨试液调pH8.3)为流动相,检测波长为280nm。[结果]峰面积对浓度的回归方程为A=21489C-20287,盐酸多西环素在20~120μg/ml浓度范围内,呈良好的线性关系(r=0.9999),平均回收率为100.84%,RSD为0.80%(n=5)。[结论]该法简便、准确,适用于盐酸多西环素片剂的含量的测定。
[Objective] An HPLC method was established to determine doxycycline hydrochloride. [Methods] The liquid chromatography method was developed, using Alhima C18 column (4.6mm×250mm, 10μm), with detection wavelength at 280 nm, mobile phase: ammonium oxalate solution (0.05 mol/L) -N, N-dimetbyl formamide-ammonium monohydrie phosphate solution (0.2 mol/L )(65 : 30 : 5). The pH value of the mobile phase is adjusted at 8.3 with ammonia. [Results] The regression equation of peak area and concentration was A=21 489 C-20 287. The linearity is good (r = 0.999 9) in the range of 20-120μg/ml. The recovery rate was 100.84%, RSD=0.80% (n = 5). [Conclusion] The method is simple, accurate and is very suitable for determination of doxycycline hvdrochloride .
