摘要
目的通过对比分析正常中国人眼前极和后极部内层巩膜组织细胞外基质中Ⅰ型胶原蛋白的分布特点,探讨其分布的差异对人巩膜组织抗拉伸力的影响,为寻求病理性近视的发病机制提供客观依据。方法应用非穿透小梁切除术获取15例自愿捐献角膜的健康正常人眼的前、后极部巩膜组织(不伴有病理性近视),用Western blot的方法检测各组巩膜组织的Ⅰ型胶原蛋白,并测定各样本β-actin和Ⅰ型胶原蛋白电泳条带灰度值,以β-actin电泳条带为参照,求出各样本Ⅰ型胶原蛋白电泳条带灰度值和β-actin电泳条带灰度值比值。结果在100kD处可见Ⅰ型胶原的蛋白条带,正常人眼后极部Ⅰ型胶原蛋白的表达水平明显低于前极部,差异有显著统计学意义(P<0.01)。结论正常人眼前极部和后极部Ⅰ型胶原纤维蛋白的分布存在差异,后极部明显低于前极部。
Objective To analyze whether or not there is a difference in the expression of collagen type I scleral extracellular matrix tissue at the anterior and posterior poles and to investigate the influence of seleral tissue in resisting the force of elongation in order to explore objective reasons for pathological myopia pathogenesis. Methods Trabeculectomy was used to obtain scleral tissue samples at the anterior and posterior poles of 15 healthy volunteers (without pathological myopia) who had donated their corneas. The expression of collagen type I was detected by Western blot, recording the IOD of β-actin and collagen type I in accordance with the electrophoresis strips' gray scale values of β-actin and collagen type I and the ratios of the two (collagen type I/β-actin), Results The protein band of collagen type I was 100 KD. There was a significant difference between the two groups (P〈0.01), Conclusion There was a significant difference in scleral tissue at the posterior and anterior poles in the normal eye. The expression of collagen type I was significantly lower at the posterior pole compared to the anterior pole.
